dc.description.abstract Çalışma, ülkemizde çay yetiştiriciliği yapılan Karadeniz Bölgesindeki farklı illerden toplanan 18 çay (Camelliya sinensis L.) genotipi ve 6 çeşidin genetik çeşitliliğinin tespit edilmesi amacı ile yapılmıştır. Çay genotiplerinin CTAB yöntemine göre DNA izolasyonu yapılmıştır. 59 adet ISSR primeri ile tüm DNA örnekleri için PCR yapılmıştır. Bu primerler içerisinden çay genotipleri arasında genetik ilişkiyi belirlemek amacıyla polimorfik olarak belirlenen 15 adet ISSR primeri kullanılmıştır. PCR işlemleri sonucunda oluşan bantlardan polimorfizm oranı %77 belirlenmiştir. PCR çalışmaları ile elde edilen ürünlerin analizi sonucu ISSR primerde 109 adet bant elde edilmiştir Elde edilen bantlar var (1) ve yok (0) şeklinde skor edilerek bunların dosyaları oluşturulmuştur. Elde edilen bant sonuçlarına göre genetik ilişki dendrogramı oluşturulmuştur. Oluşturulan dendrograma göre çay genotipleri arasında yüksek benzerlik gösterdikleri tespit edilmiştir. Çalışma sonucunda Karadeniz Bölgesine ait çay genotiplerinin sahip olduğu genetik çeşitlilik moleküler markörler ile ortaya konulmuştur.,The aim of this study was to determine the genetic diversity of 18 tea (Camellia sinensis L.) genotypes and 6 varieties collected from different provinces in the Black Sea region of Turkey. DNA isolation was performed according to the CTAB method of tea gonotips. PCR was performed for all DNA samples with 59 ISSR primers. In order to determine the genetic relationship between tea genotypes among these primers, 15 polymorphic ISSR primers were used. At the end of the PCR procedures, the polymorphism rate was determined as 77% from the bands formed. As a result of the analysis of products obtained by PCR studies, 109 bands were obtained in ISSR polymer. The obtained bands were scored as (1) and none (0) and their files were created. Genetic dendrogram relation dendongram was formed according to the obtained band results. Genetic diversity of tea genotypes of the Black Sea region was determined by molecular markers. en_US
