Acanthamoeba türleri toprak, hava ve su ortamlarında yaşar. Granülomatöz Amibik
Ensefalit (GAE) ve Acanthamoeba keratiti (AK) gibi önemli hastalıklara sebep olur.
Acanthamoeba enfeksiyonuna karşı hala etkili bir tedavi bulunmadığı için önemli bir halk
sağlığı sorunu olmaya devam etmektedir.
Çalışmada Samsun ilinden alınan su örneklerinde Acanthamoeba türlerinin
genotiplendirilmesi ilk defa yapılmıştır. Samsun ilinde belirlenen 32 istasyondan (Samsun,
Terme, Çarşamba, Tekkeköy, Bafra) alınan 192 su örneğinin 98’inde PCR ile Acanthamoeba
spp.’nin varlığı tespit edilmiştir. İstasyonlar arasında Terme ilçesinin en yüksek oranda
(%64.4) Acanthamoeba türlerinin kistleriyle bulaşlı olduğu gözlenmiştir. Bunu sırasıyla
Samsun merkez (%41.7), Çarşamba (%40), Tekkeköy (%38.9) ve Bafra (%33.3) ilçeleri
takip etmiştir. Sekans analizi sonucunda pozitif su örneklerinin 5’i Haplotip I (A.
triangularis), 29’u Haplotip II (A. polyphaga), 61’i Haplotip III (Acanthamoeba sp.) ve 3’ü
Haplotip IV olarak belirlenmiştir. Tüm istasyonlar değerlendirildiğinde araştırma alanında
en yaygın Acanthamoeba genotipinin T4 (Acanthamoeba sp., A. polyphaga, A. triangularis)
olduğu ve %3 oranında olsada T5 genotipine de rastlanıldığı belirlenmiştir.
Çalışmamızda araştırılan sularda en fazla T4 genotipine rastlanması AK’nin bölgede
yaşayan ve buradaki sularla temas halinde olan insanlar ve hayvanlar için risk
oluşturabileceği ihtimalini düşündürmektedir. Karadeniz Bölgesi’nin yanısıra Türkiye’ de
daha önce hiç çalışılmayan farklı istasyonlarda daha fazla sayıda su örneklerinde
Acanthamoeba suşlarının çeşitliliği, bunların prevalansı, genotiplendirilmesi ve patojenik
potansiyeli ile ilgili çalışmaların yapılması gerektiği kanısına varılmıştır.
Acanthamoeba species lives in air, soil and aquatic environments. Granulomatous Amoebic
Encephalitis (GAE) and Acanthamoeba keratitis (AK), are caused by Acanthamoeba spp.
Acanthamoebiasis is still one of the important public health problem, because it is not an
effective treatment against Acanthamoeba infections.
In the study, genotyping of Acanthamoeba species in water samples collected from Samsun
province was performed for the first time. The presence of Acanthamoeba spp. was detected
by PCR in 98 of 192 water samples collected from 32 sites of Samsun province (Samsun
center, Terme, Çarşamba, Tekkeköy, Bafra). Among the investigated sites, it was observed
that the highest rate of Terme was found to be contaminated by cysts of Acanthamoeba
species. This contamination was followed by Samsun center (41.7%), Çarşamba (40%),
Tekkeköy (38.9%) and Bafra districts (33.3%), respectively. As a result of the sequence
analysis, 5, 29, 61 and 3 of the positive water samples, respectively were identified as
Haplotip I (A. triangularis), Haplotip II (A. polyphaga), Haplotip III (Acanthamoeba sp.)
and Haplotip IV. When all sites were evaluated, it was determined that the most common
Acanthamoeba genotype was T4 (Acanthamoeba sp., A. polyphaga, A. triangularis) in the
investigated area and T5 genotype was found to be at the rate of 3% as well.
Our study suggests that because the T4 genotype found the most common in the investigated
waters, the AK could possible a risk to humans and animals living in the area in contact with
the waters. It has been concluded that the diversity of Acanthamoeba strains, their
prevalence, genotyping and pathogenic potential studies should be studied in more water
samples of both in the Black Sea area and Turkey.
