dc.description.abstract2015-2016 yıllarında yürütülen bu çalışma, kiraz için anaç adayı olabilecek 3 adet kiraz, 3 adet vişne, 3 adet mahlep genotipi ile Gisela 6 ve SL 64 anaçlarının in vitro koşullarda çoğaltılabilme performanslarının belirlenmesi amacıyla yürütülmüştür. Bitkisel materyal olarak kiraz, vişne, mahlep genotipleri ile Gisela 6 ve SL 64 anaçlarının sürgün uçlarından hazırlanan eksplantlar başlangıç aşamasında 0.5 mgl-1 BAP + 0.1 mgl-1 GA3 + 0.1 mgl-1 IBA içeren MS besi ortamına dikilerek kültüre alınmıştır. Enfeksiyon oranı kiraz genotiplerinde %12.03-17.87, vişne genotiplerinde %22.30-26.23 ve mahlep genotiplerinde ise %18.57-29.07 arasında gerçekleşmiştir. Sürgün çoğaltma aşamasında farklı BAP (0, 0.5 ve 1 mgl-1) dozlarının etkisi belirlenmiştir. BAP dozunun artması ile sürgün sayısının arttığı belirlenirken, en fazla sürgün sayısı 1 mgl-1 BAP dozundan elde edilmiştir. Sürgün sayısı kiraz genotiplerinde 2.14-4.10, vişne genotiplerinde 2.73-3.09 ve mahlep genotiplerinde ise 1.97-2.55 adet olarak belirlenmiştir. Köklendirme aşamasında ½ MS besi ortamındaki farklı IBA (0, 0.5, 1 ve 2 mgl-1) dozlarının köklenme oranına etkisi incelenmiştir. IBA dozunun artması ile köklenme oranının arttığı tespit edilmiş olup en fazla köklenme oranı kiraz ve mahlep genotiplerinde 2 mgl-1 IBA, vişne genotiplerinde ise en fazla 1 mgl-1 IBA dozundan elde edilmiştir. Köklenme oranı kiraz genotiplerinde %89.44-99.17, vişne genotiplerinde %76.46-96.33 ve mahlep genotiplerinde ise %29.71-63.25 olarak tespit edilmiştir.,This study, which was conducted in 2015-2016, was carried out in order to determine the propagation performance in in vitro conditions of 3 cherries, 3 sour cherries, 3 mahaleb genotypes and Gisela 6 and SL 64 rootstocks which could be the rootstock candidate for cherry. The explants prepared from the shoot tips of cherry, sour cherry, mahlep genotypes with Gisela 6 and SL 64 rootstocks as plant material were cultured in the initial stage by planting on MS nutrient medium containing 0.5 mgl-1 BAP + 0.1 mgl-1 GA3 + 0.1 mgl-1 IBA. The infection rate was 12.03-17.87 % in cherry genotypes, 22.30-26.23 % in sour cherry genotypes and 18.57-29.07 % in mahaleb genotypes. The effect of different BAP doses (0, 0.5 and 1 mgl-1) was determined in the shoot propagation stage. While increasing the number of shoots with the increase of BAP dose is determined, the maximum number of shoots was obtained from 1 mgl-1 BAP dose. The number of shoots was determined as 2.14-4.10 in cherry genotypes, 2.73-3.09 in sour cherry genotypes, 1.97-2.55 in mahaleb genotypes. The effect of different doses of IBA (0, 0.5, 1 and 2 mgl-1) on the rooting ratio of ½ MS in rooting medium was investigated. It was determined that the rooting rate increased with the increase of IBA dose, and the highest rooting rate were obtained 2 mgl-1 IBA dose in cherry and mahlep genotypes and 1 mgl-1 IBA dose in sour cherry genotypes.
