dc.description.abstractDeltametrin (DEL) ve thiakloprid (THİA) karışımı, tarımda yaygın olarak kullanılan etkili bir insektisit formülasyonudur. Bu tip insektisitlerin; heterozigotluk kaybı, gen mutasyonları ve akciğer kanserinin gelişiminde potansiyel moleküler biyogöstergelerden olan düzensiz promotor gen metilasyonu gibi moleküler değişimlere yol açabildiği ileri sürülmektedir. Ayrıca, in vitro ve in vivo araştırmalarda DEL’in reaktif oksijen türleri ve serbest radikallerin oluşumunu artırdığı ve antioksidan enzim seviyelerinin düşmesine yol açtığı bildirilmiştir. THİA’in de dahil olduğu neonikotinoit sınıfından insektisitlerin zebra balığı, fare ve sıçanda oksidatif strese ve DNA hasarına sebebiyet verdiği rapor edilmiştir. Reaktif oksijen türleri ve serbest radikaller ile antioksidanlar arasındaki dengenin bozulduğu oksidatif strese maruz kalan hücrelerde DNA ve genom etkilenmektedir, bu şekilde hücre DNA’sının yapısını değiştirebilen ROS genotoksisiteyi tetikleyerek kanser öncüsü olarak iş görür. Bu şekilde etkilenmiş hücrede, hücre bölünmesinin ve yeni hücrelerde normal kromozom dağılımının yanı sıra DNA kırıklarının sebep olduğu kromozom morfoloji değişimlerinin önlenemediği durumlarda; genomik kararsızlık, neoplastik transformasyon ve kanser gelişimi gözlenebilmektedir. Çalışmamızda kullanılması planlanan karışım halindeki DEL ve THİA insektisit kombinasyonunun insan akciğer fibroblast hücrelerinde (hTERT WHTBF-6) 24, 48 ve 72 saatlik uygulamaları sonucu hücre canlılığına ve oksidatif strese etkisi incelenmiştir. İnsektisit karışımının sitotoksik etkisi, canlı ve prolifere hücrelerin tespitini sağlayan, mitokondriyal aktivite bazlı MTT testi ile, oksidatif stres üzerindeki etkileri ise hücrelerin redükte glutatyon (GSH) seviyeleri ve hücrelerde reaktif oksijen türlerinin oluşumunu artıran lipit peroksidasyonunun bir ürünü olan malondialdehit (MDA) seviyelerinin ölçülmesiyle belirlenmiştir. İnsektisit karışımının deneyde kullanılacak dozlarının belirlenmesi ve hTERT WHTBF-6 hücre hattı için IC50 değerinin belirlenmesi amacıyla 0.50+7.5, 1.25+18.75, 2.50+37.5, 5+75, 12.5+187.5, 25+375, 50+750, 100+1500, 150+2250, 200+3000 μM’lık (sırasıyla DEL+THİA) 10 dozluk bir skala ile mtt testi gerçekleştirilmiştir. Bu deneyler sonucunda, 72 saat ugulamada IC50 değerleri deltametrin için 32 μM ve thiakloprid için 481 μM olarak belirlenmiştir. Ayrıca bu testler sonucu belirlenen insektisit karışımının 2.50+37.5, 5+75, 12.5+187.5, 25+375 μM’lık 4 dozu mtt, gsh ve mda testlerinde kullanılmıştır. Bu testler WHTBF-6 insan III akciğer fibroblastlarında S9 karaciğer fraksiyonunun varlığında ve yokluğunda uygulanmıştır. Deney verileri, deltametrin ve thiakloprid insektisit karışımlarının WHTBF-6 hücre hattında ve kullanılan deney şartlarında sitotoksik etki gösterdiğini ayrıca insektisit karışımının malondialdehit miktarını artırdığını ve glutatyon seviyesini düşürdüğünü, böylece hücrelerin oksidatif strese maruz kaldığını ortaya koymuştur.,The mixture of deltamethrin (DEL) and thiacloprid (THIA) is an effective insecticide formulation commonly used in agriculture. It's been claimed that these kinds of insecticide can lead to molecular changes such as loss of heterozygosity, gene mutations, and irregular gene methylation of the promoter, a potential molecular biomarker in the development of lung cancer. In addition, it has been reported that in vitro, and in vivo studies, DEL increases the formation of reactive oxygen species and free radicals and causes decreasing on antioxidant enzyme levels. Insecticides, the Neonicotinoid class that Thiacloprid is involved in, have been reported to cause oxidative stress and DNA damage in zebrafish, mice, and rats. In the cells, exposed to oxidative stress where the balance is disrupted between reactive oxygen species and free radicals and antioxidants, DNA and genome are affected, thereby ROS acts as a cancer precursor by triggering genotoxicity, which can alter the structure of the DNA. In such an affected cell, in case cell division and normal chromosome distribution in new cells as well as chromosome morphology changes caused by DNA fractures cannot be prevented, genomic instability, neoplastic transformation, and cancer development can be observed. The effects on cell viability and oxidative stress of DEL and THIA insecticide combination which are planned to be used in our study were investigated after 24, 48, and 72 hours of administration in human lung fibroblast cells (hTERT WHTBF- 6). While the effects of the insecticide mixture on oxidative stress were determined by measuring the reduced glutathione (GSH) levels of the cells and the malondialdehyde (MDA) levels, a product of lipid peroxidation, which increased the formation of reactive oxygen species in the cells, the cytotoxic effect of the insecticide mixture was determined by the mitochondrial activity-based MTT test, which enables the detection of viable and proliferating cells. In order to determine the doses of the insecticide mixture to be used in the experiments and the IC50 value for the hTERT WHTBF-6 cell line in 72 hours of administration, the MTT tests were performed with a 10-dose scale, 0.50+7.5, 1.25+18.75, 2.50+37.5, 5+75, 12.5+187.5, 25+375, 50+750, 100+1500, 150+2250, 200+3000 μM (DEL + THIA, respectively). As a result of these experiments, IC50 V values of 72 hours of exposure were specified as 32 μM for DEL and 481 μM for THIA. Besides, MTT, GSH, and MDA methods were also carried out with 4 doses of the insecticide mixture (2.50 + 37.5, 5 + 75, 12.5 + 187.5, 25 + 375 μM) determined by applying MTT test. These three tests had been performed in the presence and absence of S9 liver fraction in WHTBF-6 human lung fibroblasts. Experimental data showed that Deltamethrin+Thiacloprid insecticide mixtures exhibited cytotoxic effect in WHTBF-6 cell line under the experimental conditions used and that also the insecticide mixture increased the amount of malondialdehyde and decreased the level of glutathione so that the cells were exposed to oxidative stress.en_US
