Abstract: Kimyasal olarak sentezlenen ve bir vinil monomeri olan akrilamid (ACR), endüstriyel ve ticari olarak çeşitli alanlarda sıklıkla kullanılmaktadır. Endüstriyel kullanımının yanı sıra, ACR’nin yüksek sıcaklıklarda hazırlanan gıdalarda oluştuğu ve sigara dumanında bulunduğu deneysel birçok çalışmada gösterilmiştir. Bu nedenle ACR’nin insanlar üzerindeki olası sitotoksik, genotoksik ve karsinojenik etkilerinin değerlendirilmesi büyük önem arz etmektedir. ACR ile ilgili bazı çalışmalar kanser oluşumu ile pozitif ilişki kurarken, bazıları ise kanser oluşumu için ACR’nin vücuda alım şeklinin önemli olduğu şeklindeki görüşleri desteklemiştir. ACR maruziyeti ile akciğer kanseri riski arasındaki ilişki tam olarak bilinmemektedir. ACR’nin insana ait akciğer hücrelerindeki sitotoksik, genotoksik ve karsinojenik etkileri ilk kez bizim çalışmamız ile araştırılmıştır. ACR’nin sitotoksisitesi MTT testi ile; genotoksisitesi γ-H2AX ve 53BP1 kolokalizasyon testi ve Comet testi ile belirlenmiştir. ACR’nin karsinojenitesi de, neoplastik transformasyon testi (kontak inhibisyon testi) ve soft agar koloni oluşum testi (ankraj bağımsız büyüme testi) ile değerlendirilmiştir. ACR’nin deneylerde kullanılacak konsantrasyonlarının ve BEAS-2B hücre hattı için IC50 değerinin belirlenmesi amacıyla hazırlanan 10, 5, 1, 0.5, 0.1, 0.05, 0.01, 0.005 mM’lık ACR konsantrasyonları 24, 48, 72 saatlik maruziyet sürelerinde hücrelere uygulanmış ve MTT testi gerçekleştirilmiştir. 1 mM’lık konsantrasyona kadarki muamelelerde düşük bir sitotoksisite gözlenirken, 1 mM’dan itibaren sitotoksisitenin artmaya başladığı ve test edilen en yüksek konsantrasyonlar olan 5 ve 10 mM konsantrasyonlarda ise sitotoksisitede ciddi artışlar olduğu tespit edilmiştir. Uygulanacak deneyler için kullanılacak ACR konsantrasyonları 0.5, 1 ve 2 mM olarak belirlenmiştir. ACR’nin 24 saatlik maruziyetinde ko-lokalize olan odaklar hariç, γ-H2AX ve 53BP1 odaklarındaki artışların konsantrasyon artışına bağlı olduğu görülmüştür. 48 ve 72 saatlik uygulamalar sonucu ise, hem γ-H2AX, hem 53BP1 hem de ko-lokalize odak sayıları konsantrasyon artışına bağlı olarak artmıştır. ACR’nin tüm konsantrasyonlarında ve muamele sürelerinde kuyruktaki DNA yüzdesi, kuyruk momenti ve olive kuyruk momenti değerlerinde artışlar tespit edilmiştir. Neoplastik transformasyon testinde, 72 saatlik ACR muamelesini takiben BEAS-2B III hücrelerinde negatif kontrol ile kıyaslandığında çeşitli morfolojik değişikliklerin meydana geldiği görülmüştür. Soft agar koloni oluşum testinde, en yüksek koloni sayısı 2 mM’lık konsantrasyonda saptanmıştır. Sonuçlara göre 72 saatlik ACR uygulaması sonrasında, transformasyona uğrayan ve soft agarda büyüyen kolonilerin sayısında doza bağlı bir artış gözlemlenmiştir. Sonuçlarımız ACR’nin akciğer bronşial epitel hücreleri üzerinde sitotoksik etkilere ve DNA hasarına yol açabileceğini göstermiştir. Çalışmamız ACR maruziyetinin, hücrelerin morfolojilerindeki değişimleri, DNA çift zincir kırıklarının oluşumunu, proliferasyonun ve transformasyonun artışını ve transforme hücrelerin ankorajdan bağımsız bölünme ve büyüme yeteneği kazanmalarını indüklediği açıkça görülmektedir. Bu nedenle bu çalışma sonuçları, ACR maruziyetinin akciğer hücrelerinde karsinogenezi indükleyebileceği ve akciğer kanseri oluşumu açısından bir risk olabileceği hipotezini desteklemektedir.
Acrylamide (ACR), a vinyl monomer, is chemically synthesized and frequently used in various industrial and commercial areas. In addition to its industrial use, it has been shown in many experimental studies that ACR occurs in foods prepared at high temperatures and is in cigarette smoke. Therefore, evaluation of the possible cytotoxic, genotoxic and carcinogenic effects of ACR on humans is very important. While some studies about ACR have established a positive relationship with cancer formation, the others have supported the routes for ACR to enter the body is important for cancer formation. The relationship between ACR exposure and lung cancer risk is not fully known. The cytotoxic, genotoxic, and carcinogenic effects of ACR in human lung cells are investigated for the first time in our study. Cytotoxicity of ACR was determined by MTT test, and its genotoxicity was determined by γ-H2AX and 53BP1 co-localization test and comet Test. The carcinogenicity of ACR was also evaluated by the neoplastic transformation test (contact inhibition test) and soft agar colony formation test (anchorage independent growth test). ACR concentrations of 10, 5, 1, 0.5, 0.1, 0.05, 0.01, and 0.005 mM were prepared to determine the concentrations of ACR to be used in the further experiments and the IC50 values for the BEAS-2B cell line. The cells were treated with the different ACR concentrations for 24, 48 and 72 hours and then MTT test was performed. While low cytotoxicity was observed in treatments up to 1 mM concentration, it was observed that cytotoxicity started to increase from 1 mM. Significant increases in cytotoxicity were detected at 5 and 10 mM concentrations, the highest concentrations tested. The ACR concentrations to be used for the further experiments were determined as 0.5, 1, and 2 mM. It was observed concentration-dependent increases in the γ-H2AX and 53BP1 foci, except for the co-localized foci for 24 hour ACR treatment. γ-H2AX, 53BP1 and co-localized foci numbers also increased in a concentration-dependent manner for 48 and 72 hours. We found increases in the percentage of DNA in the tail, tail moment, and olive tail moment values at all ACR concentrations and treatment times. Various morphological changes were observed in BEAS-2B cells compared to V the negative control after 72 hours of ACR treatment. In the soft agar colony formation test, the highest number of colonies was detected at the 2 mM. According to the results, after 72 hours of ACR treatment, dose-dependent increases were determined in the number of transformed colonies growing on soft agar. Our results indicated that ACR can show cytotoxic effects and cause DNA damage on lung bronchial epithelial cells. We clearly showed that ACR can induce changes in the cell morphology, formation of DNA double-strand breaks, increases in cell proliferation and transformation, and the ability to divide and grow independently of the anchorage. Therefore, the results of the present study support the hypothesis that ACR exposure may induce carcinogenesis in lung cells and may be a risk for lung cancer formation.
