Abstract: Bu çalışma Yeşilırmak Deltası’ndan Temmuz-Ekim (2018) ayları arasında toplanan 3 farklı yüzen yapraklı su bitkisinin (Spirodela polyrhiza, Lemna trisulca, Salvinia natans) biyokimyasal kompozisyonu, antimikrobiyal, antioksidan, asetilkolinesteraz, bütirilkolinesteraz ve tirozinaz inhibitör aktivitelerinin belirlenmesi amacıyla yapılmıştır. Bitkilerin biyokimyasal içeriklerinin belirlenmesi için pH analizi, % kuru madde analizi, toplam azot ve protein analizi, klorofil tayini, mikro-makroelement analizi ve GC-MS analizleri yapılmıştır. Çalışmalarda bitkilerin antimikrobiyal etkilerinin belirlenmesi için etanol ve aseton ekstreleri kullanılmıştır. Bitki ekstreleri 4 Gram-pozitif, 4 Gram-negatif ve 2 mantar suşuna karşı antibiyogram ve MİK (Minimum İnhibisyon Konsantrasyonu) testleriyle test edilmiştir. Sonuçlara istatistiksel analizler uygulanmıştır. Bitkilerin antioksidan etkisinin belirlenmesi için toplam fenolik madde içeriğinin belirlenmesi için serbest radikal süpürücü aktivitesi DPPH (2,2-difenil-1-pikrilhidrazil) ve FRAP (demir iyonu indirgeyici aktivite potansiyeli) yöntemi, diğer biyokimyasal aktivitelerinin tespiti için toplam fenolik madde, asetilkolinesteraz, butirilkolinesteraz ve antitirozinaz aktiviteleri test edilmiştir. Analizler sonucunda; bitkilerin pH değerleri 6.96 (L. trisulca) ile 7.41 (S. natans) arasında tespit edilmiştir. En yüksek fotosentetik pigment değerleri (yaş ağırlıkta) S. polyrhiza türünde kaydedilmiştir (klorofil-a: 0.299 mg/g, klorofil-b: 0.108 mg/g, toplam klorofil: 0.407 mg/g ve karotenoit: 0.084 mg/g). S. natans türünde klorofil-a 0.147 mg/g, klorofil-b 0.058 mg/g, toplam klorofil 0.205 mg/g ve karotenoit değeri 0.042 mg/g’dır. Elde edilen sonuçlara göre çalışılan türler içinde en düşük pigment içeriğine S. natans türü sahiptir. % Kuru madde miktarı en fazla L. trisulca türünde (%7.10) en az ise S. natans türünde (%4.45) tespit edilmiştir. Azot ve protein sonuçlarına göre en yüksek azot (%5.71) ve protein (%35.68) miktarı S. polyhriza türünde kaydedilmiştir. En düşük azot (%3.38) ve protein (%21.12) miktarı ise L. trisulca türünde kaydedilmiştir. Antimikrobiyal analiz sonuçlarına göre etanol ekstrelerinin aseton ekstrelerinden daha yüksek etkiye sahip olduğu görülmüştür. En düşük antimikrobiyal etkiyi L. trisulca’nın aseton ekstresi verirken, en yüksek etkiyi S. polyrhiza’nın etanol ekstresi göstermiştir. S. polyrhiza’nın etanol ekstresi diğer bitkilerin de etanol ekstrelerinden daha yüksek etkiye sahiptir. Antimikrobiyal analizler sonucu, en fazla etkilenen mikroorganizma grubu Gram-pozitif bakteriler, en az etkilenenler ise funguslardır. MİK değerleri tüm ekstrelerde en düşük 81.25 μg/μL olup en yüksek değer ≥750 μg/μL’dir. Antioksidan sonuçlarına göre, en düşük DPPH radikal süpürme aktiviteleri (SC50 değeri) 0.011 mg/mL (S. polyrhiza) olarak belirlenmiştir. En düşük antioksidan etkiyi S. natans türünün aseton ekstresi göstermiştir. FRAP metodunda da en yüksek antioksidan aktiviteyi S. polyrhiza gösterirken, en düşük antioksidan aktiviteyi L. trisulca’nın aseton ekstresi vermiştir. Toplam fenolik madde, asetilkolinesteraz ve butirilkolinesteraz analizinde en yüksek değer S. polyrhiza türünde kaydedilmiştir. Asetilkolinesteraz analizinde en düşük değer L. trisulca’nın etanol ekstresinde, toplam fenolik madde ve butirilkolinesteraz analizinde ise L. trisulca’nın aseton ekstresinde tespit edilmiştir.,
This study was carried out to determine biochemical composition, antimicrobial properties, and chemical activities of three floating-leaf aquatic plant species (i.e., Spirodela polyrhiza, Lemna trisulca, and Salvinia natans) collected from Yeşilırmak Delta between July and October in 2018. pH analysis, dry matter analysis, total nitrogen and protein analysis, chlorophyll determination, micro-macroelement analysis, and GC-MS analysis were performed to determine the biochemical content of plant samples. Ethanol and acetone extracts were used to determine antimicrobial effects of plants. Plant extracts were tested against four Gram-positive, four Gram-negative, and two fungal strains using antibiogram and minimum inhibition concentration (MIC) tests. Test results were statistically analyzed. To determine the antioxidant effect and biochemical activities of plants, total phenolic substances, acetylcholinesterase, butyrylcholinesterase, and antithyrosinase activities were tested using DPPH (1,1-difenil-2-pikrilhidrazil) and FRAP (ferric reduction activity potential) methods. As a result of these analyses; pH values of plants were determined between 6.96 (for L. trisulca) and 7.41 (for S. natans). The highest photosynthetic pigment values (wet weight) were recorded in S. polyrhiza (chlorophyll-a: 0.299 mg/g, chlorophyll-b: 0.108 mg/g, total chlorophyll: 0.407 mg/g, and carotenoid: 0.084 mg/g). In S. natans, chlorophyll-a was 0.147 mg/g; chlorophyll-b was 0.058 mg/g; total chlorophyll was 0.205 mg/g; and carotenoid value was 0.042 mg/g. According to our results, S. natans had the lowest pigment content among the studied species. The highest amount of dry matter was determined in L. trisulca (7.10%) and the least was in S. natans (4.45%). According to nitrogen and protein results, the highest amount of nitrogen (5.71%) and protein (35.68%) were recorded in S. polyhriza. The lowest amount of nitrogen (3.38%) and protein (21.12%) were recorded in L. trisulca. According to antimicrobial analysis results, ethanol extracts had a higher effect than acetone extracts did. The lowest antimicrobial effect belonged the acetone extract of L. trisulca, while the ethanol extract of S. polyrhiza was the most effective extract. Moreover, the ethanol extract of S. polyrhiza had a higher effect than that of other plants. As a result of antimicrobial analysis, the most affected microorganism group was Gram-positive bacteria and the least affected were fungi. The lowest and highest MIC values were 81.25 and 750 μg/μL, respectively. According to antioxidant results, the lowest DPPH radical scavenging activities (SC50 value) was 0.011 mg/mL (in S. polyrhiza). The acetone extract of S. natans showed the lowest antioxidant effect. While S. polyrhiza showed the highest antioxidant activity according to FRAP method, acetone extract of L. trisulca had the lowest antioxidant activity. The highest value was recorded in S. polyrhiza in the analysis of total phenolic substances, acetylcholinesterase, and butyrylcholinesterase. In the acetylcholinesterase analysis, the lowest value was determined in the ethanol extract of L. trisulca. In total phenolic substance and butyrylcholinesterase analysis, acetone extract of L. trisulca had the lowest value.
