Abstract: Mantarlar, yıllardır gıda maddesi olarak tüketilmekte ve nefis lezzetleri, ekonomik ve ekolojik değerleri ve tıbbi özellikleri açısından takdir edilmektedir. Mantarların, protein, karbonhidrat, ham lif, vitamin, mineral ve ham yağ kaynakları olduğu bilinmektedir. Yenilebilir mantarlar ayrıca, fenolik bileşikler, steroller ve triterpenler gibi biyoaktif ikincil metabolitler de içerir. Bu besinler ve metabolitler, yenilebilir mantarlar için rapor edilen biyolojik etkinliklerden sorumludur. Bu bilgiler ışığında, bu tez çalışmasında, Ordu ilinin farklı kesimlerinden toplanan yabani ve yenilebilir Cantharellus cibarius, Lactarius deliciosus ve Lactarius pyrogalus mantar türlerinin biyolojik olarak aktif madde profilinin belirlenmesi amaçlandı. Bu amaçla, metanol ile ekstrakte edilen mantar örneklerinin, FolinCiocalteu yöntemi ile toplam fenolik içerikleri gallik asit eşdeğeri olarak 5.338- 2.272 mg/g numune arasında hesaplanırken, toplam flavonoid miktarı ise AlCl3 metodu ile kursetin eşdeğeri olarak 5.020-17.540 mg/g numune değerleri arasında saptandı. Bunların dışında doğal fenoliklerden biri olan tannik asit eşdeğeri cinsinden mantar numunelerinin toplam tannin miktarları ise Folin-Denis reaktifi kullanılarak spektrofotometrik yöntemle tespit edildi ve en yüksek değer 7.160 mg/g numune olarak L. deliciosus için gözlemlendi. Numunelerin kurutulmuş metanol ekstraktları kullanılarak, askorbik asit, karoten ve likopen içerikleri de hesaplanmış olup elde edilen tüm bulgular, incelenen tüm mantar numunelerinin etkin ölçüde biyoaktif bileşen içerdiğini öngörmektedir. Öte yandan, bu biyoaktif bileşenlerin türünü aydınlatabilmek için mantar numunelerimizin ekstraktları, HPLC ile fenolik bileşen analizine tabii tutuldu ve test edilen 18 tür fenolik bileşiğin 12 tanesinin incelenen mantar numunelerinde mevcut olduğu görüldü. Bu fenolik bileşiklerin miktarca göze çarpanları pirogallol, benzoik asit ve resveratrol olarak sayılabilir. Elde edilen bulgular test edilen mantar numunelerinin literatürdeki diğer pek çok çalışma ile uyumlu olabilecek şekilde gıda olarak kullanıldığında, vücudumuzdaki antioksidatif bileşen ihtiyacını karşılayabilecek düzeyde sekonder metabolit içerdiğini ortaya koymaktadır. Bu tez çalışması ile elde edilen bulgular gıda, kozmetik, ilaç ve tıp alanlarında yararlanılabilecek bir kaynak olarak literatüre önemli bir katkı sağlamakla birlikte benzer çalışmalara ışık olabilecek niteliktedir.,
Mushrooms have been consumed as food and appreciated for their exquisite flavor, economic and ecological values, and medicinal properties for many years. Mushrooms are known that they are sources of protein, carbohydrate, crude fiber, vitamin, mineral and crude fat. Edible mushrooms also contain bioactive secondary metabolites such as phenolic compounds, sterols and triterpenes. These nutrients and metabolites are responsible for the biological activities reported for edible mushrooms. In the light of this information, it is aimed to determine the biologically active substance profile of wild and edible Cantharellus cibarius, Lactarius deliciosus and Lactarius pyrogalus mushroom species collected from different parts of Ordu province in present thesis study. For this purpose, the total phenolic contents of the mushroom samples extracted with methanol by Folin-Ciocalteu method were calculated between 5.338- 2.272 mg/g sample as gallic acid equivalent, while total flavonoid amounts were detected between 5.020-17.540 mg/g sample as quercetin equivalent using AlCl3 method. In addition, the total amount of tannin in the mushroom samples of the tannic acid equivalent, which is one of the natural phenolics, was determined spectrophotometrically using Folin-Denis reagent and the highest value was observed for L. deliciosus as 7.160 mg/g sample. Ascorbic acid, carotene and lycopene contents were also calculated using dried methanol extracts of the samples, and the all obtained data suggest that all examined samples contained an effective amount of bioactive component. On the other hand, in order to illuminate the variety of these bioactive compounds, the extracts of samples were subjected to phenolic component analysis by HPLC and 12 of the 18 phenolic compounds tested were found to be present in the examined samples. Pyrogallol, benzoic acid and resveratrol among all phenolics were attracted the attention as quantitatively. The findings reveal that the tested mushroom samples contain secondary metabolites that are able to meet the antioxidative component requirements of our bodies when used as food, consistent with many other studies in the literature. Findings obtained with this thesis study can be light for similar working with providing an important contribution to the literature as a resource that can be used in food, cosmetics, drug and medicine fields.
