Bu çalışma 2017-2018 yıllarında Ordu Üniversitesi Ziraat Fakültesi Bahçe Bitkileri
Bölümü Biyoteknoloji laboratuvarında yürütülmüştür. Denemede kullanılan 35 adet
kendilenmiş ve saflaştırılmış nitelikli biber ıslah hatları ile 3 adet ticari biber çeşidi
Karadeniz Tarımsal Araştırma Enstitüsü Müdürlüğünden temin edilmiştir. Çalışmada
19 adet SRAP primer kombinasyonu kullanılarak biber hatlarının genetik
karakterizasyonu yapılmıştır. Ayrıca biber ıslah hatlarında moleküler markırlar
yardımıyla PVY, TSWV ve PMMoV’ne dayanıklılık taramaları yapılmıştır. Üç
burun biber tipine sahip G13 ıslah hattı her üç hastalığa da dayanıklı çıkmıştır. 19
SRAP markırı toplam 85 allel üretmiş ve bunların 56’sı biber hatları arasında
polimorfizm göstermiştir. Primer kombinasyonu başına ortalama polimorfik allel
sayısı 2.94 olarak tespit edilmiştir. Primer kombinasyonlarının ortalama polimorfizm
bilgi içeriği değerleri 0.17 ile 0.98 arasında değişmiş ortalama 0.58 bulunmuştur.
ME01+EM02 ile ME18+EM11 primer kombinasyonlarından en yüksek polimorfik
allel sayıları (6 allel) elde edilmiştir. ME04+EM08 primer kombinasyonunun
polimorfizm bilgi içeriği değeri de 0.98 olarak belirlenmiştir. SRAP markırları
verilerine dayalı oluşturulan dendrogramda biber ıslah hatları 5 ana gruba ayrılmıştır.
Biber hatlarının benzerlik indeks değerleri 0.35 ile 0.97 arasında değişmiştir.
Korelasyon matrisinin dengrograma yansımasını gösteren mantel test değeri r = 0.90
olarak hesaplanmıştır. Temel bileşen eksenlerinden ilk 4’ü biber hatları arasındaki
toplam varyasyonun % 84.9’unu açıklamıştır.,This study was conducted in biotechnology laboratory of Ordu University

groups. Similarity index values of pepper lines were changed between 0.35 and 0.97.
