Abstract: Bu araştırma, Yalova ili sera alanlarında yetiştiriciliği yapılan kesme çiçeklerde görülen zararlı bitki paraziti nematodların tespit edilmesi ve tespit edilen nematodların morfolojik, morfometrik ve moleküler karakterizasyonlarının belirlenmesi için gerçekleştirilmiştir. Bu amaç için 2017 yılı üretim sezonunda 6 farklı kesme çiçek çeşidi gül (Rosa spp.), şebboy (Matthiola spp.), krizantem (Chrysanthemum spp.), hüsnü yusuf (Dianthus barbatus), Lisianthus (Eustoma spp.) ve frezya (Freesia spp.) üretimi yapılan seralarda örneklemeler gerçekleştirilmiştir. Alınan örneklerden elde edilen nematodların tür teşhisleri ve yoğunlukları tespit edilmiştir. Elde edilen nematodların tür teşhisleri moleküler yöntemler kullanılarak gerçekleştirilmiştir. Moleküler analizlerde ITS ve D2/D3 gen bölgeleri ile türlere özgü spesifik primer çiftleri kullanılmıştır. Ayrıca, nematodların morfolojik karakterleri ve morfometrik ölçümleri gerçekleştirilerek incelenmiştir. Yapılan moleküler analizler sonucunda Meliodogyne incognita (Kofoid & White) Chitwood, Meliodogyne hapla (Chitwood), Aphelenchus avenae (Bastian), Aphelenchoides sp., Trophurus imperialis (Loof), Longidorus moesicus (Lamberti, Choleva and Agostinelli.), Pratylenchus vulnus (Allen and Jensen), Paratylenchus hamatus (Thorne and Allen), Rotylenchus arasbaranensis (Atighi, Pourjam, Ghaemi, Pedram, Liebanas, Cantalapiedra-Navarrete, Castillo & Palomares-Rius) türü nematodlar tespit edilmiştir. Çalışmada kesme çiçek yetiştiriciliği yapılan alanlarda saptanan en yaygın türler Aphelenchus avenae, Aphelenchoides sp., Meloidogyne incognita ve Rotylenchus arasbaranensis olarak tespit edilmiştir.,The aim of this study was to determine plant parasitic nematodes in cut flowers grown in greenhouses in Yalova province and to determine morphological, morphometric and molecular characterization of these nematodes. For this purpose, surveys were carried out in the greenhouses where 6 different cut flower varieties including rose (Rosa spp.), Tenweeks stock (Matthiola spp.), Chrysanthemum spp., hosni yusuf (Dianthus barbatus), Lisianthus (Eustoma spp.) and Freesia spp. were produced during the production season of 2017. Species identification and density of nematodes obtained from the samples were determined. Species identification of the obtained nematodes was carried out using molecular methods. ITS and D2 / D3 gene regions and specific species-specific primer pairs were used in molecular analyzes. Morphological characteristics and morphometric measurements of nematodes were also investigated. As a result of the molecular analyzes, Meliodogyne incognita (Kofoid & White) Chitwood, Meliodogyne hapla (Chitwood), Aphelenchus avenae (Bastian), Aphelenchoides sp., Trophurus imperialis (Loof), Longidorus moesicus (Lamberti, Choleva and Agostinelli), Pratylenchus vulnus (Allen and Jensen), Paratylenchus hamatus (Thorne and Allen), Rotylenchus arasbaranensis (Atighi, Pourjam, Ghaemi, Pedram, Liebanas, Cantalapiedra-Navarrete, Castillo & Palomares-Rius) species were found.
